High-Throughput Screen for Modulators of Oncogenic CARD11 Signaling
CARD11 is a multi-domain scaffold protein whose function is required for regulated NF-kappaB activation during the adaptive immune response. Several hyperactive mutations have been identified in Diffuse Large B-Cell Lymphoma (DLBCL). While some of these oncogenic mutants selectively recruit known co-factors of CARD11 such as Bcl-10, others fail to recruit these co-factors. Using a high-throughput screen, we are identifying previously unknown co-factors of oncogenic CARD11 which may modulate its activity during the adaptive immune response.
